What Is Epitalon? Telomerase and Longevity Research Explained
Epitalon is a synthetic tetrapeptide, a chain of four amino acids, studied for its effect on telomerase, the enzyme that rebuilds the protective ends of chromosomes. The founding finding comes from cultured human cells, where Khavinson et al. (2003) reported telomerase activity and telomere elongation after epitalon treatment. Later work has reported telomere extension in additional human cell lines, in a 2025 study by authors outside the compound’s developers, and rodent studies have reported longer maximum lifespan in some conditions. Human data are still limited, so many of the key questions remain open for researchers.Ā
Epitalon structure and origin
Epitalon, also spelt epithalon, has the amino acid sequence Ala-Glu-Asp-Gly, shortened to AEDG. It was developed by a Russian research group led by Vladimir Khavinson, who is a co-author on the 2003 mouse lifespan study covered below. The peptide belongs to the group of short peptides often called bioregulator peptides or Khavinson peptides.
Epitalon is modelled on epithalamin, a complex of peptides isolated from the pineal gland. Epitalon is the synthetic tetrapeptide, and the two are described as separate preparations in the research literature. Results for one therefore do not transfer automatically to the other.

How does epitalon activate telomerase?
Telomeres are repeating DNA sequences that cap the ends of chromosomes. In young human somatic cells they shorten by up to 70 base pairs a year, and cells stop dividing once telomeres become too short, according to the introduction of a 2025 Biogerontology paper. Telomerase adds telomeric repeats back. Its catalytic subunit is encoded by the TERT gene (hTERT in humans). The same paper notes that telomerase is absent from mature somatic cells but reactivated in around 90% of human cancers.
The proposed mechanism for epitalon sits at the gene level. Khavinson et al. (2003) added epitalon to telomerase-negative human fetal fibroblasts and reported induced expression of the catalytic subunit, enzymatic telomerase activity and telomere elongation. The authors proposed that this reflects reactivation of the telomerase gene in somatic cells.
The 2025 study by Al-Dulaimi and colleagues, whose authors are not among the compound’s developers, tested epitalon across several human cell types, including the breast cancer lines 21NT and BT474 and normal epithelial and fibroblast cells. The results differed by cell type. The journal has since published a correction that replaced the paper’s figures 1 to 3.
- Normal cells: dose-dependent telomere extension through upregulation of hTERT and telomerase.
- Cancer cell lines: significant telomere extension through ALT (alternative lengthening of telomeres), a telomerase-independent route. Normal cells showed only a minor increase in ALT activity.
Both studies used cultured cells, which show what a compound does in a dish. Whether the same change occurs in the tissues of a living adult has not been established.
What do animal studies show about epitalon and lifespan?
Lifespan data come mainly from rodents, and the pattern is mixed.
- Female SHR mice: Anisimov et al. (2003) gave 54 mice per group monthly courses of subcutaneous injections from three months of age. Mean lifespan did not change. Maximum lifespan rose by 12.3%, and the lifespan of the last 10% of survivors rose by 13.3%. Chromosome aberrations in bone marrow cells fell by 17.1%. Total spontaneous tumour incidence was unchanged, while leukaemia development was 6.0-fold lower.
- Female rats: in a 2007 study in Bulletin of Experimental Biology and Medicine, epitalon did not change lifespan under a standard day and night light regime. Under natural and constant light, both of which shortened lifespan in untreated rats, maximum lifespan was prolonged by 95 and 24 days respectively.
The common thread is that mean lifespan did not move under standard conditions, and the effects appeared in maximum lifespan or in animals under stress. Rodent strains and housing conditions limit how far these results travel, and the mouse study included the compound’s developers among its authors.
Epitalon, the pineal gland and melatonin
The pineal gland produces melatonin, the hormone that follows the day and night cycle. A Russian-language 2007 paper by Korkushko and colleagues reported that epithalamin and epitalon restored night-time melatonin release in old monkeys. In elderly people with reduced pineal function, night-time melatonin rose.
Rat studies point in different directions. The 2025 overview review of epitalon describes one experiment in isolated, perifused pineal glands from young and aged rats, where epitalon did not change melatonin release, including release stimulated by the drug isoproterenol. In a separate study of cultured pinealocytes (pineal cells), epitalon had a significant effect on AANAT, an enzyme in melatonin synthesis, and on pCREB, a signalling protein.
Neither result establishes an effect on sleep in people.
How strong is the evidence for epitalon in humans?
The human data cited here are the melatonin work above, which measured hormone levels in elderly volunteers, and a 2002 report in retinitis pigmentosa from the group that developed epitalon, which included both rats and patients. The telomerase findings come from cultured cells. The table sets each claim against the type of study behind it.
| Claim | Evidence level | What was reported | Main limitation | Source |
| Raises telomerase and lengthens telomeres | Cell culture (human fibroblasts and cell lines) | Telomerase expression, activity and telomere extension after treatment | Cultured cells, not living tissue. Cancer lines showed ALT activation | Khavinson 2003, Al-Dulaimi 2025 (correction) |
| Extends lifespan | Animal (mice and rats) | No change in mean lifespan. Maximum lifespan up 12.3% in female mice. Rat results depended on lighting | Rodent models. Developers among the authors of the mouse study | Anisimov 2003, 2007 rat study |
| Restores night-time melatonin | Old monkeys and elderly human volunteers | Higher night-time melatonin in old monkeys and in elderly people with reduced pineal function | Isolated rat pineal glands showed no change in melatonin release | Korkushko 2007, 2025 review |
These findings describe biological activity in the model systems tested and do not establish a clinical effect in healthy people. River Peptides supplies epitalon for in-vitro laboratory research only, and it cannot be used in human trials or administered to humans.
Purity and identity verification in epitalon research
Mass spectrometry measures a compound’s molecular mass, which researchers compare against the expected value of about 390 g/mol for epitalon. HPLC (high-performance liquid chromatography) separates the main peptide from related substances to give a purity figure. Commercial epitalon is commonly supplied as an acetate or trifluoroacetic acid (TFA) salt, according to the overview review. The counterion adds weight to the powder, so net peptide content is worth checking on a certificate of analysis.
At River Peptides, samples from each new batch go to independent laboratories, including Janoshik Laboratories in the Czech Republic and Analiza Biatek in Poland. The batch ID printed on the label links to the matching certificate of analysis. The independent testing page sets out the full process, and the Epitalon 10mg page carries its certificate of analysis. Epitalon is also available as a 10mg spray. River Peptides holds ISO 9001 and ISO 14001 accreditation, reflecting our quality management and environmental standards.Ā

Epitalon FAQs
What is epitalon used for in research?
Researchers study epitalon in cell culture and animal models of ageing. The main areas are telomerase activity and telomere length, pineal gland function and melatonin, and lifespan in rodents. It is not an approved treatment for any condition.
Is it epitalon or epithalon?
Both spellings refer to the same Ala-Glu-Asp-Gly tetrapeptide. Epitalon is the more common spelling in recent research papers, so this article uses it.
What does AEDG stand for?
AEDG strings together the one-letter codes for alanine (A), glutamic acid (E), aspartic acid (D) and glycine (G), in sequence order.
How is epitalon stored in research settings?
According to the Epitalon 10mg technical specifications, unopened lyophilised powder is kept at -20 °C with freeze-thaw cycles minimised, and its shelf life is two years under those conditions. Once reconstituted under sterile conditions, it is stable for up to 28 days at 2 to 8 °C.
