Sermorelin vs CJC-1295: Choosing a GH-Releasing Peptide for Research
Sermorelin and CJC-1295 are GHRH analogues that act on the same pituitary receptor. They differ in structure, half-life and the way they stimulate growth hormone (GH) release. Sermorelin reproduces the first 29 amino acids of human GHRH. CJC-1295 modifies that sequence to resist breakdown, and its DAC version adds a linker that binds albumin and extends activity to days.
This sermorelin vs CJC-1295 comparison sets out the research strengths and limits of each peptide and shows which suits which experimental question. For the wider picture on GHRH analogues and GHRPs, see the GH secretagogues guide.

How Sermorelin and CJC-1295 Differ in Structure
Sermorelin is a 29-amino-acid peptide that matches the amino-terminal segment of the 44-residue native hormone. It binds the GHRH receptor and stimulates GH secretion from the pituitary, according to the PubChem compound record. It serves as the reference sequence for the modified analogues below.
CJC-1295 without DAC, also called Mod GRF 1-29 or tetrasubstituted GRF(1-29), keeps the same 29-residue backbone and changes four residues. Compared with the Semoreline sequence, the substitutions are:
- Position 2: alanine becomes D-alanine
- Position 8: asparagine becomes glutamine
- Position 15: glycine becomes alanine
- Position 27: methionine becomes leucine
The D-alanine at position 2 is the change aimed at dipeptidyl peptidase-IV, the enzyme that cleaves native GRF between positions 2 and 3. In porcine plasma, a D-amino acid at that position prevented the cleavage (Su et al., 1991).
CJC-1295 With DAC vs Without DAC
DAC stands for drug affinity complex. It is a reactive linker added to the C-terminus that binds covalently to circulating albumin, which lengthens the half-life from minutes to days. The without-DAC form lacks that linker. In rats, the albumin-bound form stayed in plasma beyond 72 hours and showed greater stability against dipeptidyl peptidase-IV (JettƩ et al., 2005). The full explainer on CJC-1295 DAC covers the chemistry in more detail.
GH Release Pattern and Half-Life: Implications for Study Design
Half-life sets how long each compound stimulates the receptor, and therefore what kind of GH signal a design can produce.
- Sermorelin: 11 to 12 minutes
- CJC-1295 without DAC: longer than sermorelin, far shorter than the DAC form
- CJC-1295 with DAC: 5.8 to 8.1 days
In a placebo-controlled study of the DAC form, a single injection in healthy adults raised mean GH concentrations two- to ten-fold for six days or more, and raised IGF-1 1.5- to 3-fold for 9 to 11 days (Teichman et al., 2006). A follow-up study in healthy young men found that GH pulsatility persisted under this continuous stimulation, while trough and mean GH levels rose (Ionescu and Frohman, 2006). The DAC form therefore raises the baseline without replacing the pulses.
Three design observations follow from these profiles:
- Discrete, time-limited stimulation: sermorelin and CJC-1295 without DAC, because receptor exposure falls within a short window after each addition
- Sustained exposure over days: CJC-1295 with DAC, because a single addition covers a multi-day window
- Effect of sequence modification alone: sermorelin against CJC-1295 without DAC, because the backbone is shared and only four residues differ
What the Research Evidence Covers for Each Peptide
Sermorelin has the longer clinical history. It was formerly FDA-approved as Geref, and the FDA later determined that its discontinuation was for reasons other than safety or effectiveness, as set out in the Federal Register notice. A 1992 study in healthy older men found that twice-daily GHRH(1-29) for 14 days raised GH and IGF-1 levels.
The main published human studies of CJC-1295 are Teichman 2006 and Ionescu and Frohman 2006. Both study the DAC form in healthy adults, and both list Lawrence Frohman as an author. Neither covers the without-DAC form, so designs that rely on it draw on the shared GHRH receptor pharmacology rather than on compound-specific human work cited here.
Research Advantages of Sermorelin and CJC-1295
Sermorelin in Research
- Native reference sequence: residues 1 to 29 of GHRH give a baseline against which modified analogues can be compared
- Established record: a longer published human history than either CJC-1295 form, as covered above
- Brief stimulus: an 11 to 12 minute half-life suits designs that need short, defined receptor stimulation
The same short half-life limits designs that need sustained exposure without repeated additions to the system. River Peptides supplies Sermorelin 5mg for laboratory research, with batch-level testing. Our sermorelin overview covers the compound in more depth.
CJC-1295 Without DAC in Research
- Protease resistance: four substitutions protect the sequence from the enzymatic breakdown that clears native GHRH quickly
- Short-acting profile: a half-life far below the DAC form keeps stimulation closer to discrete pulses
- Clean comparison with sermorelin: the shared backbone isolates the effect of the substitutions
The main limit is that the human studies cited here cover the DAC form rather than this one. CJC-1295 without DAC is available in 5mg and 10mg vials.
CJC-1295 With DAC in Research
- Sustained receptor exposure: albumin binding extends activity to several days, as shown in Teichman 2006
- Pulsatility retained: GH pulses persisted during continuous stimulation in Ionescu and Frohman 2006
- Fewer additions per study window: one addition covers a period that would need many with the short-acting forms
Sustained exposure answers a different experimental question from pulsatile stimulation, so the DAC form cannot stand in for the short-acting compounds. Its long half-life also complicates washout between conditions. CJC-1295 with DAC 5mg is listed in the River Peptides range.

Sermorelin vs CJC-1295 FAQs
Is sermorelin the same as CJC-1295?
No. Both act on the GHRH receptor and share a 29-residue backbone. CJC-1295 carries four amino acid substitutions that slow breakdown, and the DAC version adds an albumin-binding linker. Sermorelin has the native sequence.
Which has the longer half-life, sermorelin or CJC-1295?
CJC-1295 with DAC, by a wide margin. Its estimated half-life is 5.8 to 8.1 days (Teichman 2006), against 11 to 12 minutes for sermorelin. CJC-1295 without DAC sits between the two.
Does sermorelin or CJC-1295 have more published research?
Sermorelin has the longer clinical history, including its former FDA approval and studies such as Corpas 1992. The main published human studies of CJC-1295 are two 2006 papers on the DAC form.
Which is better for research, sermorelin or CJC-1295?
It depends on the question. Sermorelin suits reference work and short, defined stimulation. CJC-1295 without DAC suits studies of the modified sequence. CJC-1295 with DAC suits sustained exposure designs. The decision table above maps each question to a compound.
Can sermorelin and CJC-1295 be compared in the same study design?
Yes, with care. Sermorelin and CJC-1295 without DAC share a backbone and have comparable short half-lives, which makes them a clean pair. The DAC form changes exposure over days, so comparisons with it need to account for the different time course. Find out the difference between DAC and no DAC in CJC-1295.
How is the identity and purity of sermorelin and CJC-1295 verified?
Independent laboratories test each batch. HPLC confirms purity and mass spectrometry confirms identity, and the batch ID on the label locates the matching certificate of analysis. The third-party testing section above lists the laboratories and methods.
